Scientists successfully crack the genome of diploid cotton

The international research team led by Chinese Academy of Agricultural Sciences and BGI have completed the genome sequence and analysis of a diploid cotton-- Gossypium raimondii. The cotton genome provides an invaluable resource for the study and genetic improvement of cotton quality and output, and sheds new lights on understanding the genetic characteristics and evolutionary mechanism underlying cotton and its close relatives. The study was published online in Nature Genetics.

Cotton, also known as "white gold", is an important cash crop worldwide. Its fiber is one of the oldest fibers under human cultivation, which traces over 7,000 years old recovered from archaeological sites. The cotton production provides income for approximately 100 million families, and approximately 150 countries are involved in cotton import and export. Additionally, in scientific research, cotton also serves as an excellent model system for studying polyploidization, cell elongation and cell wall biosynthesis.

In this study, researchers sequenced the genome of G. raimondii by the next-generation sequencing technology, yielding a draft cotton genome with 103.6-fold genome coverage. Over 73% of the assembled sequences were anchored on 13 G. raimondii chromosomes. They identified 2,355 syntenic blocks in the G. raimondiigenome, and found that approximately 40% of the paralogous genes were present in more than 1 block, which suggests that this cotton genome has undergone substantial chromosome rearrangement during its evolution.

Through comprehensive comparison and analysis, researchers observed that one paleohexaploidization event occurred in the G. raimondiigenome at approximately 130.8 million years ago, while the event is commonly found in eudicots. They also found the evidence to support a cotton?specific whole?genome duplication event occurred at approximately 13-20 million years ago.

Cotton is known to produce a unique group of terpenoids such as gossypol. The accumulated gossypol and related sesquiterpenoids produced by cotton in pigment glands can be as a resistance against pathogens and herbivores. The majority of cotton sesquiterpenoids are derived from a common precursor which is synthesized by (+)- ? -cadinene synthase (CDN) in gossypol biosynthesis. Through the phylogenetic analysis on G. raimondii and eight other sequenced plant genomes, they found that the cotton, and probably Theobroma cacao, were the only sequenced plant species that possess an authentic CDN1 gene family for gossypol biosynthesis.

Furthermore, the transcriptomic comparison between the fiber-bearing G. hirsutum and the non-fibered G. raimondiidemonstrated that three synthases are important for cotton fiber development, including sucrose synthase (Sus), 3-ketoacyl-CoA synthase (KCS) and 1-aminocyclopropane-1-carboxylic acid oxidase (ACO). Meanwhile, the MYB and bHLH transcription factors preferentially expressed in fiber may be useful to explain the molecular mechanisms that are in charge of governing fiber initiation and early cell growth.

Zhiwen Wang, Project Manager at BGI, said, "The completed G. raimondii genome provides a good reference for accelerating the genomic research on tetraploid cotton species such as G. hirsutum and G. barbadense. It also will lay a solid foundation for researchers to further boost cotton quality and productivity by comprehensively exploring the genetic mechanisms underlying cotton fiber initiation, gossypol biosynthesis and resistance against pathogens and herbivores."

Ill. Postman Admits Stealing Mailed Charity Donations - Philanthropy ...

A former Chicago-area mail carrier pleaded guilty Thursday to stealing nearly 30,000 letters containing donations to a charity on his route, reports the Chicago Tribune.

Frederick L. Taylor, a 12-year employee of the Berwyn, Ill., post office, admitted withholding mail containing $275,000 in checks, cash, and money orders intended for the unnamed nonprofit group between August 2010 and August 2011. He pocketed nearly $18,000 in cash gifts from the letters, authorities said.

Mr. Taylor, 41, pleaded guilty to one count of possessing stolen mail and faces up to five years in prison. Sentencing was scheduled for November 19.

Has a summer cold or mold allergy stuffed up your nose and dampened your sense of smell? We take it for granted that once our nostrils clear, our sniffers will dependably rebound and alert us to a lurking neighborhood skunk or a caramel corn shop ahead.

That dependability is no accident. It turns out the brain is working overtime behind the scenes to make sure the sense of smell is just as sharp after the nose recovers.

A new Northwestern Medicine study shows that after the human nose is experimentally blocked for one week, brain activity rapidly changes in olfactory brain regions. This change suggests the brain is compensating for the interruption of this vital sense. The brain activity returns to a normal pattern shortly after free breathing has been restored.

Previous research in animals has suggested that the olfactory system is resistant to perceptual changes following odor deprivation. This new paper focuses on humans to show how that's possible. The study is published in the journal Nature Neuroscience.

"You need ongoing sensory input in order for your brain to update smell information," said Keng Nei Wu, the lead author of the paper and a graduate student in neuroscience at Northwestern University Feinberg School of Medicine. "When your nostrils are blocked up, your brain tries to adjust to the lack of information so the system doesn't break down. The brain compensates for the lack of information so when you get your sense of smell back, it will be in good working order."

For the study, Wu completely blocked the nostrils of 14 participants for a week while they lived in a special low-odor hospital room. At night, participants were allowed to breathe normally while they slept in the room.

After the smell deprivation, researchers found an increase in activity in the orbital frontal cortex and a decrease of activity in the piriform cortex, two regions related to the sense of smell.

"These changes in the brain are instrumental in maintaining the way we smell things even after seven days of no smell," Wu said.

When unrestricted breathing was restored, people were immediately able to perceive odors. A week after the deprivation experience, the brain's response to odors had returned to pre-experimental levels, indicating that deprivation-caused changes are rapidly reversed.

Such a rapid reversal is quite different from other sensory systems, such as sight, which typically have longer-lasting effects due to deprivation. The olfactory system is more agile, Wu suggested, because smell deprivation due to viral infection or allergies is common.

This study also has clinical significance relating to upper respiratory infection and sinusitis, especially when such problems become chronic, at which point ongoing deprivation could cause more profound and lasting changes, Wu noted.

"It also implies that deprivation has a significant impact on the brain, rather than on the nose itself," Wu said. "More knowledge about how the system reacts to short-term deprivation may provide new insights into how to deal with this problem in a chronic context."

Judge grants Sheryl Crow 3-year restraining order

SANTA MONICA, Calif. (AP) ? A judge granted Sheryl Crow a three-year restraining order against a man who acknowledged he threatened to shoot the singer-songwriter and film executive Harvey Weinstein.

Philip Gordon Sparks, 45, agreed to stay away from the Grammy winner and Weinstein after an hour-long hearing in which he accused the pair of stealing $7.5 million from him, videotaping and following him without permission and leaving him homeless. A forensic psychiatrist who interviewed Sparks recently called him "imminently dangerous" and said his psychosis is directed intently at Crow.

Superior Court Judge James Hahn ordered Sparks to stay 300 yards away from Crow and Weinstein and make no attempt to contact them.

Hahn, the former Los Angeles mayor who was appointed as a judge in 2008, presided over the hour-long hearing, patiently instructing Sparks to ask questions and allow him to describe at length his reasons for feeling persecuted.

Crow obtained a temporary restraining order against the 45-year-old in July after her team reviewed a series of Sparks' online posts directed at the singer.

A worker at the Screen Actors Guild-American Federation of Television and Radio Artists wrote in a declaration accompanying Crow's filing that she spoke with Sparks on July 16 and he made the threat against Crow. He also threatened to shoot film executive Weinstein because he believed they were filming him and had stolen millions from him, the worker stated.

Crow, 50, has won nine Grammy Awards and has two sons, ages 5 and 2, who are also covered by the order.

Sparks said he made the statement because he was frustrated because he believes they stole from him and continued to follow him.

"Mr. Sparks is unambiguously delusional," forensic psychiatrist Dr. David Glaser testified during the hearing.

Neither Crow nor Weinstein attended the hearing.

New York - City Comptroller John C. Liu today called on Mayor Bloomberg to reform NYPD policy to allow Sikh officers to serve without having to forsake their turbans and beards.

In a letter, Comptroller Liu commended the Mayor for his words of comfort and support to the City?s Sikh community in the wake of the Wisconsin shootings, saying that now is the time for New York City to make a meaningful change of inclusion to Sikh members of the NYPD.

?The attack in Wisconsin reminds us that intolerance still rears its ugly head today. Sikhs have felt alienation and distress over the past decade since September 11. In the wake of the Wisconsin killings, which have shocked the national conscience, our City can enact meaningful inclusion of this community by changing NYPD rules to allow Sikhs to serve without having to forsake their turbans and beards,? Comptroller Liu wrote.

Comptroller Liu noted that the Washington D.C., Metro Police Department has amended its rules to allow Sikh officers to wear religious garb and pointed to a recent City Council law that requires workplaces to foster supportive environments for religious practices.

Comptroller Liu said that by reforming the NYPD policy, individuals of other faith groups who wear head coverings and grow beards, as required by their religion, would also benefit.

?Shouldn?t the NYPD, providing security in one of the most diverse cities in the world, proudly display its own diversity?? Comptroller Liu said.

Jasminder Kaur, project manager of United Sikhs, said:

?It?s imperative for people to know that Sikhs are woven into the fabric of American society and are not the ?other.? One way to promote that is through increasing participation of Sikhs with turbans and beards in government jobs. We appreciate Comptroller Liu for taking the initiative to bring this up with the NYPD and assuring us of his continuous support.?

Harpreet Singh Toor, Chairman of Public and External Affairs for the Sikh Cultural Society, said:

?It is heartening to see that all of America came out to support the Sikh community in this time of tragedy, but as long as the systematic discrimination continues at the city, state, and federal levels, Sikhs will always feel themselves to be lesser Americans than others. Rabbi David Zwiebel, Executive Vice President of Agudath Israel of America, said:

Attorney: Husband acted out of love in hospital killing

An Ohio man accused of fatally shooting his wife of 45 years in a hospital intensive care unit acted out of love, his attorney and a longtime co-worker said on Wednesday.

According to police, John Wise was armed with a gun when he went to the?neuroscience ICU at Akron General Medical Center in Akron, Ohio, and shot Barbara Wise, 65, in the head. A doctor who was also in the room at the time wasn't injured.?

Wise?s attorney, Paul Adamson, says Wise acted out of mercy.

"I'm thoroughly convinced he's a good man. I think his past history bears that out," Adamson told the Cleveland Plain Dealer after a court appearance on Wednesday.

"Forty-five years of marriage, blessed to be deeply in love with his wife throughout those 45 years, and I am absolutely confident that everything that he's ever done for his wife has been done out of deep love, including the events that just recently transpired."

Terry Henderson, 58, who said he worked with John Wise, of Massillon, Ohio, for 30 years, told The Associated Press that he believes Wise wanted to spare his wife from suffering when he shot her Saturday in her hospital bed. She died the next morning.

Barbara Wise had been admitted to the hospital last week with a non-disclosed, life-threatening condition.

Husband charged in fatal hospital shooting of wife: 'Is she not dead?'

Henderson said Wise, 66, was a good husband without a hint of domestic violence. He said Wise never wanted to become disabled in a nursing home and that Barbara Wise felt the same way.?

Wise appeared before an Akron municipal court judge via video from jail Wednesday morning for hi arraignment. The original attempted murder charge against him was upgraded to aggravated murder after results of Barbara Wise's autopsy.?

No plea was entered. Bond was set at $1 million. Wise must return to court Aug. 22.

Work on Fiji?s first casino will begin this month as investors will meet to give contracts to local companies in Denarau this week.

One Hundred Sands ? the company behind the project has managed to raise $400million for construction.

However the initial projections for phase one of the project which will see the construction of a convention center, casino and resort is $290m.

Managing Director Larry Claunch says overseas investors are often given misconceptions about Fiji?s investment potential by local private sectors.

?There has been a lot of criticism if we could raise the money in a timely fashion ? we have, criticism and comments if we can finish on time ? we will ? its going to be probably a fastest development anybody has ever seen and it going to be fun to watch.?

Claunch says the current government played a vital role for this investment.

?I didn?t come to Fiji with a though of developing a casino or anything for that matter ? I really just came here to eat fresh fish ? but after seeing the vision of this government I knew everything was possible and I was all in.?

Five hundred locals will be employed during the construction and around 600 to 850 will be employed at the casino.

The new casino is expected to begin operation from September next year.

Aspers Casino looks forward to end of Games...

Though it?s happening right on the doorstep of his company?s flagship casino, Richard Noble is looking forward to when the 2012 London ?Olympic Games finish.

The chief operating officer of Aspers Group ? a joint venture between the Australian Securities Exchange-listed Crown and the Aspinall family, which operates Aspers Casino at Westfield Stratford City next to Olympic Park ? says weekly takings and average visitor numbers are down about 20 per cent in the first week of the Games.

When Aspers at Westfield Stratford City, the country?s largest casino, opened on December 1 last year, Noble told The Australian Financial Review it was immediately popular. ?In our first three days we had 30,000 visitors which was about 100 per cent above our business case. After that we were averaging 25,000-30,000 visitors per week. But in the last few months we?ve seen the three carparks here at the Westfield get cut down to one and now it is none, so we have been adversely affected.?

There have been restrictions placed on visitor numbers to Westfield Stratford by the London Olympic Games Organising Committee (LOCOG). These include only Games ticket holders being allowed access to Westfield between 8.30am and 5pm on Friday and Saturday.

Noble says this cut down on the ability of regular Aspers patrons to visit the casino. He said it also affecting the numbers of people in the local area attracted to Westfield during the Games, who could have potentially visited the casino for the first time. LOCOG is also likely to institute another similar ?lock-out? later this year due to fears about congestion at the mall, which is between Olympic Park and public transport links.

With the Olympics not providing the boost in business once hoped, Noble says Aspers will now concentrate its marketing efforts on the post-Games period. ?On the 18th of ?September, after the Olympics and then the Paralympics are over, we will be having a huge relaunch.?

Noble says the casino concentrates on a high volume of customers who spend relatively little. It has a catchment area of 4.1 million people.

?And that is just in the east and the area around, north to Luton and east to Southend,? he said.

?But what?s been surprising and also delightful is that we?ve managed to get a lot of visitors from the west of London as well.?

A change to British gaming laws in 2007 allowed for the casino to be built without previous restrictions that meant patrons had to become members. Aspers also owns casinos in Newcastle, Swansea and Northampton and is planning to open a new venue in Milton Keynes in 2003.

Overall, he says Aspers is pleased about how the Stratford business has performed given it has already expanded since its December 2011 opening. It now has 57 gaming tables, the highest number in Britain, up from 38 when it opened. It has room to accommodate 80 tables. There are also now 138 electronic gaming terminals, up from 92. The site will soon feature a bespoke poker room to go with the sports bar, VIP bar and terrace, which has been hired out to corporate clients for the Games.

But Noble, who is otherwise enjoying the Games, can?t wait until September. ?Some of our patrons are not able to get here at the moment but that will change once all this is over.? (The Australian Financial Review)

Police detain 150 in Macau casino raids amid fears of new gang war...

HONG KONG (Reuters) - Police detained more than 150 people in weekend raids on casinos and hotels in Macau after a recent spate of killings raised fears of a new gang war in the world's largest gambling destination.

The former Portuguese territory was gripped by gang wars in the late 1990s and the recent murder of three people in two weeks, thought linked to the gambling industry, comes just months ahead of the release from jail of triad boss "Broken Tooth" Wan Kuok-koi, who wreaked havoc at that time.

About 1,300 people were rounded up on Friday and Saturday in Macau and 150 of those were taken in for questioning in the operation codenamed "Thunderbolt".

"This operation is an annual exercise to maintain public security. It is a joint exercise undertaken by police in Guangdong province, Macau and Hong Kong to crack down and prevent crime in this region," a Macau police spokeswoman said.

In Hong Kong, police raided 21 locations on August 2 as part of the joint operation and arrested 130 people on suspicion of various crimes including money laundering involving HK$300 million (24.77 million pounds), illegal gambling and prostitution.

Police also seized cash, watches and cars worth more than HK$11 million.

The Macau raids came just weeks after a Chinese woman was found murdered in a residential area minutes away from the cavernous gambling halls of gaming magnate Sheldon Adelson's Venetian casino.

That followed the murders of two Chinese nationals at the five-star Grand Lapa hotel in Macau, a one-hour ferry ride from Hong Kong, and an attack at the end of June on a senior figure in Macau's junket industry, which extends credit to rich gamblers.

By contrast, only five homicide cases were recorded between June 2011 and May 2012, according to Macau police statistics.

Macau, which like Hong Kong is a special administrative region of China, has boomed since the 1999 handover from Portugal, with Las Vegas moguls including Adelson and Steve Wynn setting up glitzy casino hotels.

That growth, however, has slowed significantly in the past three months, forcing junket operators into more aggressive debt-collecting tactics.

Many of the smaller junket companies, which collect gambling debts in exchange for a commission from casino operators, are struggling to stay in business.

American computerised gaming equipment designer and manufacturer International Game Technology (IGT) has signed a deal that will see the Golden Acorn Casino and Travel Center in California host an online casino application from its Double Down Interactive subsidiary.

Las Vegas-based IGT purchased the casual games developer in January and revealed that the Golden Acorn Casino is set to utilise its technology in order to offer casino players free-play gaming experiences.

July appeared to be a Stephen King month for me. Most of the short fiction I read was King and every novel I read was King as well. Short fiction still suffered this month, but that is in part because I read three fairly long books (two of the three books added up to nearly 2,000 pages!) and I went on an article reading binge mid-month as well. I have a backlog of short fiction to catch up on. If only I can find the time to do it!

Incidentally, Stephen King?s?It (my second reading of it) marks my 494th book since January 1, 1996. I?ll likely finish book #495 today and since I have 2 books to read for my September book review column, that puts me at least 497 books by the end of August. Anyone want to venture a guess as to when I will hit book #500?

Alaska GOP's ?old guard? questions what happened to ?new guard? funds

Frank McQueary, assistant treasurer of the Republican Party of Alaska, sent an email to GOP party officials on Monday about ?potentially improper fundraising by Chair Elect Russ Millette.?

McQueary wrote that as assistant treasurer of the party, along with his role as standing rules chair, he has had ?numerous enquiries forwarded to me regarding both interpretations of the rules and questions concerning the financial aspects of fundraising.?

Millette was not immediately available for comment.

McQueary told the group that he had contacted the Alaska Public Offices Commission (APOC), the state agency that monitors elections, seeking an opinion on whether funds have been properly reported by Millette. ?

At issue is more than $1,600 raised by the splinter group, or ad hoc committee, that met June 9 in a church after the reconvening of the Republican Party Convention, McQueary said.

?To the best of my belief, the funds in question were not turned over to the party for reporting purposes, nor have they been reported by any other organization,? McQueary wrote.?

Money raised by any political party must be documened. The APOC indicated it?s investigating the issue.

McQueary said that party practice dictates that donor information is received for money raised. If the donor is unknown, the party sends the money to the state Department of Revenue.

Funds were requested at various times before and during the meeting. Some of that money was used, according to a transcript of the meeting, to pay for a parliamentarian the group had hired from Washington state.

At the end of the meeting, Millette told the group that it had collected $1624.60. ?This covers expenses for (the parliamentarian), which includes his airfare,? Millette told the group. He also said that another person in the crowd, Gene Brokaw, was paying for the church and that members could pay him back.

The business manager of the church confirmed that it had charged the group $200 to use the building.?

The June 9 reconvening was the brainchild of Millette, Alaska?s next chair of the Alaska GOP. He put out a call for state Republicans to finish what they started during the?tumultuous state GOP convention?in late April.?Millette is a supporter of GOP presidential candidate Ron Paul.?

With the help of failed 2010 U.S. Senate candidate Joe Miller, Millette captured the hearts and minds of Alaska's Paul supporters and dissatisfied Republicans to win the state GOP chairmanship at the April convention.

The war between the two factions of Alaska?s GOP has brewed since. At the June 9 reconvening, the group voted to censure U.S. Sen. Lisa Murkowski for joining ?far left interest groups? and ?maliciously attacking? Joe Miller, who ran as a Tea Party candidate in the 2010 U.S. Senate race, but lost in the general election to Murkowski's write-in campaign.

Censuring didn?t stop with Murkowski. The group also voted to censure a handful of Republican Party officials who weren?t in the room, including McQueary.

Millette will become chair of the party early next year. In the meantime, he is the treasurer of the party, and old party boss Randy Ruedrich is still in charge.?
